Abstract:

The purposes of the present study were to screen the ability of Aspergillus fumigatus to produce antibacterial compounds using different growth parameters namely, pH, temperature, agitation and time of fermentation and investigate the effect of the yield against bacterial isolates. A. fumigatus was isolated from arable lands in Sudan and identified macroscopically and microscopically. The pure fungal culture was inoculated in fermentation medium with different growth parameters. The fungal metabolites were extracted in intervals of 7, 8, and 9 days incubation period. No yield was observed at pH values of 6 and 8. A relatively high yield of active compounds was observed at pH 7, agitation speed of 75 rpm and 7 days contact time, while the highest yield was exhibited at pH 7, agitation speed of 100 rpm and 8 days contact time. These metabolites were investigated for their inhibitory effects against indicator bacterial species. All indicator bacterial species showed resistance to 2.5% concentration of the crude extract. The 5% crude extract concentration exhibited small inhibitory effect (8 mm) against Salmonella typhimurium, moderate inhibitory effect (19 mm) against Listeria monocytogenes and high inhibitory effect (25 mm) against Pseudomonas aeruginosa. The pronounced inhibitory effect was observed with 10% concentration against Pseudomonas aeruginosa (40 mm) followed by Listeria monocytogenes (30 mm). A moderate inhibitory effect of the 10% crude extract was observed with Salmonella typhimurium (13 mm). The indicator bacteria Staphylococcus aureus showed resistance to all tested concentrations. The present study show that the growth parameters affect the yield but not the viability of Aspergillus fumigatus. The fungal metabolites have antibacterial effects.

Production and characterization of biochemical properties of L-Asparaginase by indigenous yeast isolated from soil of Iran

Introduction: The use of L-asparaginase as a chemotherapeutic agent is an effective strategy for the treatment of leukemic lymphoblasts. Since the use of bacterial L-asparaginase causes side effects due to hypersensitivity reactions, search for new enzyme sources is one of the active research areas. In this study, the characterization of produced L-asparaginase by indigenous yeast strains was i...

Pectinase and Polygalacturonase Production by a Thermophilic Aspergillus Fumigatus Isolated from Decomposting Orange Peels

A thermophilic fungal strain producing both pectinase and polygalacturonase was isolated after primary screening of 120 different isolates. The fungus was identified as Aspergillus fumigatus Fres. MTCC 4163. Using solid-state cultivation, the optimum levels of variables for pectinase and polygalacturonase (PG) production were determined. Maximal levels of enzyme activities were achieved upon gr...

Optimization of Cellulase Production from Bacteria Isolated from Soil

Cellulase-producing bacteria were isolated from soil and identified as Pseudomonas fluorescens, Bacillus subtilIs, E. coli, and Serratia marcescens. Optimization of the fermentation medium for maximum cellulase production was carried out. The culture conditions like pH, temperature, carbon sources, and nitrogen sources were optimized. The optimum conditions found for cellulase production were 4...

Production and Characterization of Exopolysaccharide Produced by Alcaligenes Faecalis B14 Isolated from Indigenous Soil

Exo-polysaccharides (EPS) are environment friendly natural polymers secreted by microorganisms in the surrounding medium. Due to the presence of unique structural composition, EPS shows diverse applications such as in food formulations, pharmaceutical, and cement based construction industry, etc. In the present investigation, the bacteria producing higher exopolysaccharide was screened among ba...

Production of Antibacterial Compounds from Actinomycetes

Actinomycetes are soil-dwelling Gram-positive bacteria, industrially relevant as producers of a wide range of bioactive secondary metabolites, including many anti‐ biotics of clinical and commercial importance. The understanding of actinomycete biology has been based on extensive studies on the model organism Streptomyces coelicolor over many years and on the availability of its complete genome...

Induction and Optimization of Chitosanase Production by Aspergillus fumigatus YT-1 Using Response Surface Methodology

In recent years, much attention has been paid to chitosan oligosaccharides (COS) because COS have shown physiological activities such as antitumor effect and antimicrobial activity.1 Compared with chitosan, chitosan oligosaccharides (COS) have some advantages such as water solubility, lower viscosities, lower molecular weights and shorter chain lengths. COS could be obtained from chitosan hydro...
